e17623 Background: Endometrial cancer (EC) is one of the most prevalent gynecologic tumors. Current diagnosis and treatment EC no longer rely solely on traditional histopathological classification. Nevertheless, molecular classification demonstrated clear prognostic value may guide clinical decision. Methods: In this study, archived tissue specimens from 240 patients Department Pathology, Peking University People’s Hospital. Four subtypes [POLE ultramutated (POLE mut), microsatellite instability high (MSI-H), copy number low (CNL), (CNH)] were stratified by next-generation sequencing (NGS) panel (Amoy Diagnostics, Xiamen, China) targeting POLE, TP53, BRCA1, BRCA2 genes (MSI) status. Immunohistochemistry (IHC) was applied to detect expression P53, MMR other related proteins. Results: Distribution in 13 (5.42%) POLE mut, 36 (15.00%) MSI-H, 41 (17.08%) CNH, 150 (62.50%) CNL. Compared published results Caucasian including TCGA, ProMisE as well TransPORTEC, real-world data Chinese ECs displayed a significantly larger proportion addition, novel biomarkers such DUSP1, MCF7 BUB1, which independent marker our previous research validated. Also, it found that appeared be more than BRCA1. Further analysis revealed overall consistency for NGS-based IHC-based TP53 abnormalities detection MSI/MMR status assessment 87.5% 100%, respectively. Conclusions: have unique characteristics. order perform accurate typing ECs, indicators match characteristics population need added existing classifiers. easy operate replicate with accuracy. Thus, viable alternative IHC

Molecular pathogenesis of endometrial cancer
Introduction. Genetic changes which cause endometrial cancer may be discovered by simple comparison of tumor with normal tissues, and correlating clinicopathologic with genotypic features. More difficult is the specific elaboration of those interacting events which may transpire in a particular sequence during a protracted interval of carcinogenesis. A small burden of premalignant cells makes t...

We comprehensively evaluated genetic variants in the thymidylate synthase (TYMS) gene in association with endometrial cancer risk in a population-based case-control study of 1,199 incident endometrial cancer cases and 1,212 age frequency-matched population controls. Exposure information was obtained via in-person interview, and DNA samples (blood or buccal cell) were collected. Genotyping of 11...
